Description

Description: This assay employs a two-site sandwich ELISA to quantitate XIRP1 in samples. An antibody specific for XIRP1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any XIRP1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for XIRP1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of XIRP1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: The deduced chicken and mouse proteins contain 2,562 and 1,677 amino acids, respectively. Both have an N-terminal DNA-binding domain, followed by a repeated 16-amino acid module, 1 or 2 SH3-binding motifs, and nuclear localization signals.
Xin exhibited a peripheral localization within cardiomyocytes and remained at cell-cell junctions throughout development and into adulthood. Colocalization of Xin with beta-catenin (CTNNB1) and N-cadherin (CDH2), in addition to coimmunoprecipitation experiments, suggested that Xin localized to the adherens junctions within the intercalated disc. At embryonic day 10.0, Xin was also found within developing somites and eventually at the myotendon junctions of skeletal muscle
